Antibody Immunogen Source: Human TBC1D7 sequence peptide Cat. No.: PE-04AEE99
Antibody Immunogen Sequence: SVY(pY)EKV(bA)C
Antibody Immunogen Description: Corresponds to amino acid residues S11 to V17; in the N-terminus of protein. The effect of Y14 phosphorylation is unclear. This is the main phosphosite targeted by the ERK1 and ERK2 MAP kinases. This is the major sites of in vivo phosphorylation of TBC1D7 based on ≥88 mass spectrometry reports recorded in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.
Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.
Antibody Concentration: 1 mg/ml
Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol
Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.
Product Use: Western blotting | Antibody microarray
Antibody Dilution Recommended: 2 µg/ml for immunoblotting
Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Dog | Rat | Mouse | Platypus | Frog | Zebra fish | Fruit fly.
Antibody Positive Control: The observed molecular mass of the processed target protein on SDS-PAGE gels is reported to be around 25-34 kDa.

Scientific Background: TBC1D7 (TBC1 domain family member 7) is a ~34-kDa protein that acts as the essential third core subunit of the tuberous sclerosis complex (TSC1-TSC2-TBC1D7), which inhibits mTORC1, a master regulator of cell growth. It possesses GTPase-activating protein (GAP) activity toward Rheb, a small GTPase that activates mTORC1, and its depletion results in increased mTORC1 signalling. TBC1D7 stabilizes the TSC1-TSC2 complex (also known as the hamartin-tuberin complex) by binding to TSC1. The protein is stabilized by AKT, and its association with the TSC complex is regulated by growth factors. It is widely expressed, with high expression observed in the human heart. TBC1D7 is found in the cytoplasm, often localizing to the endomembrane system within the TSC complex. Mutations in the TBC1D7 gene are linked to autosomal recessive megalencephaly (abnormal enlargement of the cerebral hemispheres), intellectual disability, and large head size (macrocephaly).
