Antibody Host Species: Rabbit
Antibody Immunogen Source: Human snRNP 70 sequence peptide Cat. No.: PE-04AFF99
Antibody Immunogen Sequence: FEV(pY)GPI(bA)C
Antibody Immunogen Description: Corresponds to amino acid residues F123 to I129; in the RRM_1 domain. The effect of Y126 phosphorylation is unclear. This is the major sites of in vivo phosphorylation of snRNP70 based on ≥643 mass spectrometry reports recorded in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris. This antibody was also subject to negative purification over phosphotyrosine-agarose.
Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Scientific Background: SNRNP70 is part of the spliceosomal U1 snRNP, which is essential for recognition of the pre-mRNA 5' splice-site and spliceosome assembly. It is a critical RNA-binding protein that acts as a core subunit of the U1 small nuclear ribonucleoprotein (U1 snRNP) complex, initiating pre-mRNA splicing by recognizing the 5' splice site. SNRNP70 binds to the loop I region of U1-snRNA. It features an RNA recognition motif (RRM), an α-helical domain, and arginine/serine-rich (RS) domains that facilitate interactions with other splicing factors (e.g., SRSF1). SNRNP70 is primarily located in the nucleoplasm, though it is found in the cytoplasm during certain phases. It is linked to autoimmune diseases such as mixed connective tissue disease (MCTD) and systemic lupus erythematosus (where it acts as an autoantigen). It forms detergent-insoluble aggregates in Alzheimer's disease and co-localizes with Tau, making it a biomarker for the condition.
