Antibody Immunogen Description: Corresponds to amino acid residues V87 to L93; in the Sit domain. Y90 phosphorylation induces interaction with Grb2. This is the major in vivo phosphorylation site of SIT based on ≥337 mass spectrometry reports recored in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SIT (Signaling Threshold-Regulating Transmembrane Adapter Protein) negatively regulates TCR (T-cell antigen receptor)-mediated signalling in T-cells. It is involved in positive selection of T-cells. SIT-deficient mice showed that the absence of SIT leads to T-cell hyperresponsiveness and higher production of T-helper 1 (TH1) cytokines. It contains tyrosine-based signaling motifs (TBSMs) in its cytoplasmic tail. When phosphorylated, these motifs allow SIT to recruit inhibitory molecules (such as the phosphatase SHP-2) to the TCR complex, dampening activation signals. SIT helps set the activation threshold during T-cell selection in the thymus, preventing premature activation. Unlike many other adaptors found in lipid rafts, SIT is largely excluded from the lipid raft fraction of the plasma membrane. It plays a role in preventing autoimmunity; SIT-deficient mice display increased susceptibility to developing experimental autoimmune encephalomyelitis.
