Antibody Immunogen Description: Corresponds to amino acid residues P780 to A786; in the intervening region between the second SH2 domain and the SH3 domain in PLCG1. Y783 phosphorylation stimulates phospholipase activity, induces interaction with Grb2 and VEGFR-2 and regulates cell motility. This is one of the major sites of in vivo phosphorylation of PLCG1 (≥566 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). PLCG1 is known to be phosphorylated at this site in vitro by EGFR, Flt1 (VEGFR1), ITK, Ret, Src, Syk, TrkB (NTRK2), and ZAP70.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: PLCG1 mediates the production of the second messenger molecules diacylglycerol (DAG) and 1D-myo-inositol 1,4,5-trisphosphate (IP3) from the hydrolysis of 1-phosphatidyl-1D-myo-inositol 4,5-bisphosphate. It becomes activated in response to ligand-mediated activation of receptor-type receptor-tyrosine kinases, such as PDGFRA, PDGFRB, FGFR1, FGFR2, FGFR3 and FGFR4. By virtue of its SH2 domains, PLCG1 binds to tyrosine-phosphorylates receptors and become activated by direct tyrosine phosphorylation by these receptors.
