Antibody Immunogen Description: Corresponds to amino acid residues E965 to V971; in the insert region that separates the SH3_2 and SH3_1 domains. The effect of Y968 phosphorylation is unclear. This is the second major in vivo phosphorylation site in ITSN2 (≥765 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). Phosphorylation is increased in reponse to EGF.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ITSN2 is an adapter protein that may provide indirect link between the endocytic membrane traffic and the actin assembly machinery. It acts as a key coordinator of clathrin-mediated endocytosis, cytoskeletal remodeling, and signal transduction. It coordinates vesicle maturation and budding by interacting with proteins like Eps15, Dab2, and dynamin, specifically facilitating the internalization of receptors such as EGF and T-cell receptors. It regulates the formation of clathrin-coated vesicles (CCPs), and seems to be involved in CCPs maturation including invagination or budding. ITSN2 is involved in endocytosis of integrin beta-1 (ITGB1) and transferrin receptor (TFR). It also plays a role in dendrite formation by melanocytes. ITSN2 is a multimodular protein existing in short and long isoforms due to alternative splicing. The Short Isoform contains two Eps15 homology (EH) domains, a coiled-coil region, and five SH3 domains (SH3A-E). The Long Isoform features the short isoform structure plus a RhoGEF domain (DH/PH) (acts as a specific guanine nucleotide exchange factor for Cdc42) and a C2 domain. ITSN2 is expressed ubiquitously, co-localizing with Eps15 and other endocytic machinery components, with specific isoforms showing tissue-specific expression (e.g., isoform 1 in heart/liver) and localization within the cytoplasm and at the centrosome.
