Antibody Immunogen Description: Corresponds to amino acid residues L819 to L825; in the fourth C2 domain. The effect of Y822 phosphorylation is unclear. This is the major in vivo phosphorylation site in ESYT1 (≥1233 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ESYT1 (Extended Synaptotagmin-1) is an endoplasmic reticulum (ER)-anchored protein that binds glycerophospholipids in a barrel-like domain and may play a role in cellular lipid transport. It is anchored to the ER via an N-terminal hairpin, with a SMP (synaptotagmin-like, mitochondrial, and lipid-binding) domain and multiple C2 domains that bind phospholipids and calcium. It translocates to sites of contact between the endoplasmic reticulum and the cell membrane in response to increased cytosolic calcium levels. It helps to tether the endoplasmic reticulum to the cell membrane and promotes the formation of appositions between the endoplasmic reticulum and the cell membrane. It has also been found to localize at ER-mitochondria contact sites, and plays a role in regulating mitochondrial lipid transport and respiration.
