Antibody Immunogen Description: Corresponds to amino acid residues E223 to M229; in the N-terminal portion of EML4 near the HELP domain. The effect of Y226 phosphorylation is unclear. This is the major in vivo phosphorylation site in EML4 (≥408 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: EML4 is a microtubule-binding protein that may modify the assembly dynamics of microtubules, such that microtubules are slightly longer, but more dynamic. It is essential for microtubule stability and helps organize the mitotic spindle during cell division. It is involved in proper kinetochore attachment to microtubules and promotes the recruitment of NUDC to the mitotic spindle. EML4 features a tandem atypical propeller domain (TAPE), a hydrophobic motif (HELP), a basic region, and a trimerization domain (TD). It is widely expressed in human tissues. The N-terminal region of EML4 fuses with the kinase domain of ALK (Anaplastic Lymphoma Kinase). The EML4 coiled-coil domain (part of the TD) facilitates constitutive dimerization of the fusion protein, leading to constant, oncogenic activation of ALK. The EML4-ALK fusion protein is a potent oncogene found in 4-5% of non-small cell lung cancers (NSCLC), often in young, non-smoking adenocarcinoma patients.
