Antibody Immunogen Description: Corresponds to amino acid residues L138 to G144; in the middle of the GTP_EFTU domain. The effect of Y141 phosphorylation is unclear. This is the second major in vivo phosphorylation site in EEF1A1 (≥1206 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: EEF1A1 (Eukaryotic Translation Elongation Factor 1 Alpha 1) is an abundant translation protein that promotes the GTP-dependent binding of aminoacyl-tRNA to the A-site of ribosomes during protein biosynthesis. Beyond translation, it acts as a multifunctional protein involved in heat shock response, actin bundling, and apoptosis regulation. It forms a complex with PARP1 and TXK that acts as a T helper 1 (Th1) cell-specific transcription factor and binds the promoter of IFN-gamma to directly regulate its transcription, and is thus involved importantly in Th1 cytokine production. It is primarily found in the cytoplasm, with significant presence in the nucleus. It is widely expressed in tissues like the liver, lung, and pancreas. Its paralog, EEF1A2 (with 97% amino acid similarity), replaces it in adult brain, heart, and skeletal muscle.
