Antibody Immunogen Description: Corresponds to amino acid residues Y235 to L241; in the third Annexin domain. The effect of Y238 phosphorylation is unclear. This is the second major in vivo phosphoryation site in Annexin A2 with 964 mass spectrometry reports recorded in PhosphoSitePlus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: Annexin A2 (ANXA2) is a calcium-regulated membrane-binding protein that binds two calcium ions with high affinity, which is greatly enhanced by anionic phospholipids. It may be involved in heat-stress response and inhibits PCSK9-enhanced LDL receptor degradation. It regulates membrane trafficking, endocytosis, exocytosis, and actin remodeling. It operates as a monomer in the cytoplasm or as a heterotetramer (with S100A10) on the cell surface to promote fibrinolysis, angiogenesis, and cell signaling. ANXA2 is predominantly located in the cytosol and on the cytoplasmic face of the plasma membrane, but can exist in the nucleus. It is upregulated in various cancers, including breast, liver, and prostate cancer, and facilitates tumour growth and metastasis.
