Antibody Immunogen Description: Corresponds to amino acid residues S243 to F249; in the second CH domain. The effect of Y246 phosphorylation is unclear. This is the major in vivo phosphorylation site in ACTN1 from mass spectrometry studies (≥528 reports in PhosphoSite Plus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

It acts as an antiparallel homodimer, forming bundles of filaments involved in cell structure and spreading. It is a 95-kDa, dumbbell-shaped protein containing an actin-binding domain and a Ca2+-dependent regulatory domain (EF-hands). It crosslinks actin filaments, specifically managing cortical actin reorganization in endothelial cells. It is located along microfilament bundles and in focal adhesions, facilitating cell motility, lamellipodial dynamics, and the organization of focal adhesions. Genetic variants in ACTN1 cause congenital macrothrombocytopenia, a condition characterized by low levels of abnormally large platelets. Increased expression of ACTNA has been linked to poor survival in osteosarcoma and enhanced cell invasion/migration in other cancers, making it a potential diagnostic marker.
