Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: IQGAP2 (IQ motif-containing GTPase-activating protein 2) is a multidomain scaffolding protein that acts as a tumour suppressor and regulator of cell morphology, motility, and inflammatory responses. It binds F-actin, calmodulin, and Rho GTPases (Rac1/Cdc42; but not increasing their GTPase activity), acting primarily in the liver, where it counters oncogenic IQGAP1 and regulates metabolism, and suppresses liver tumorigenesis. It contains a calponin homology domain (CHD), WW domain, four IQ motifs (binding calmodulin), and a RasGAP-related domain. It is needed for the inflammatory response in the colon (via NF-κB/IL-6) and impacts blood-brain barrier integrity. Reduced expression of IQGAP2 is associated with severe liver, hepatocellular carcinoma, breast, gastric, and thyroid cancers, as well as Alzheimer’s disease. Loss of IQGAP2 in breast and gastric cancers promotes epithelial-mesenchymal transition (EMT) and cancer progression. Isoform 2 expression is enhanced in testis.
