Product Type Specific: IQGAP1 phosphosite-specific antibody
Antibody Code: PN1029
Antibody Target Type: Phosphosite-specific
Antibody Phosphosite: S1443
Protein UniProt: P46940
Protein SigNET: IQGAP1
Antibody Type: Polyclonal
Antibody Host Species: Rabbit
Antibody Immunogen Source: Synthetic phosphopeptide patterned after human IQGAP1
Antibody Immunogen Sequence: KSK(pS)VKED(bA)C
Antibody Immunogen Description: Corresponds to amino acid residues K1440 to D1447.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.
Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.
Antibody Concentration: 0.3 mg/ml
Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol
Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.
Product Use: Western blotting | Antibody microarray
Antibody Dilution Recommended: 2 µg/ml for immunoblotting
Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human.

Scientific Background: IQGAP1 (IQ motif-containing GTPase-activating protein 1) is a 190 kDa scaffolding protein that acts as a central hub for regulating cytoskeletal architecture, cell-cell adhesion, and signaling pathways. It facilitates diverse cellular functions—including cell migration, polarization, and proliferation—by binding to over 90 partners, including activated Cdc42/Rac1, F-actin, Actin-related protein 2/3 (Arp2/3) complex, APC, E-cadherin and beta-catenin, calmodulin, ERK1/2 and Raf kinases . It is composed of five main domains: a calponin-homology domain (CHD) for actin binding, WW domain, IQ-motif domain (for calmodulin binding), Ras GAP-related domain (GRD), and a Ras GAP C-terminal domain (RGCT). It does not activate the GTPase activity of Cdc42 or Rac1. It is a critical mediator in MAPK/ERK and PI3K/Akt signaling cascades, often promoting cell survival. It directly binds and stabilizes actin filaments and tethers them to microtubules, promoting cell motility and controlling cell shape. It is highly expressed in the placenta, lung, and kidney, with lower levels seen in the heart, liver, skeletal muscle and pancreas. It is often overexpressed in tumours (e.g., breast, liver, glioma), IQGAP1 acts as a potential oncogene, driving cell proliferation and migration.
