Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: HOMER1 (Homer protein homolog 1) is a postsynaptic density (PSD) scaffolding protein of the homer family that is critical for regulating excitatory synapses, glutamate receptor signaling, and calcium homeostasis. It plays a key role in neuronal plasticity, learning, and memory. It acts as an adaptor protein that binds and cross-links cytoplasmic regions of GRM1, GRM5, ITPR1, DNM3, RYR1, RYR2, SHANK1 and SHANK3. By physically linking GRM1 and GRM5 with ER-associated ITPR1 receptors, it aids the coupling of surface receptors to intracellular calcium release. It may also couple GRM1 to PI3 kinase through its interaction with AGAP2. It regulates the maintenance of dendritic spines and the scaling of synaptic AMPA receptors. It occurs in different isoforms.
