Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CTMP (Acyl-coenzyme A thioesterase; THEM4) is a cytosolic and mitochondrial enzyme that belongs to the THEM4/THEM5 thioesterase family. It exhibits acyl-CoA thioesterase activity towards medium and long-chain (C14 to C18) fatty acyl-CoA substrates, and probably plays a role in mitochondrial fatty acid metabolism. It catalyzes the reaction: hexadecanoyl-CoA + H2O = hexadecanoate + CoA + H+. It functions in the apoptotic process, possibly via its regulation of protein kinase B (PKB/Akt) activity and location. It binds to the regulatory domain of Akt and inhibits its phosphorylation, acting as a crucial modulator of cell survival, proliferation, and apoptosis. It is found predominantly in skeletal muscle, testis, uterus, brain and kidney. Its is down-regulated in glioblastoma or glioma compared to non-neoplastic brain due to promoter hypermethylation.
