Antibody Immunogen Description: Corresponds to amino acid residues D342 to S349. Phosphorylation of T346 and S348 stimulate the enzymatic activity of LASS2. These are among the major in vivo phosphorylation sites based on ≥258 and ≥217 mass spectrometry reports, respectively recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: LASS2 (LAG1 longevity assurance homolog 2; Ceramide Synthase 2; CerS2) is a ubiquitously expressed enzyme that that catalyzes the transfer of the acyl chain from acyl-CoA to a sphingoid base, with high selectivity toward very-long-chain fatty acyl-CoA (chain length C22-C27). The ceramide synthetase catalyzes the reaction: a very long-chain fatty acyl-CoA + a sphingoid base => an N-(very-long-chain fatty acyl)-sphingoid base + CoA + H+. LASS2 influences cell behavior by interacting with V-ATPase (regulating pH and proton pumps) and regulation of ferroptosis signaling through TFRC. It also promotes p53 stability and can enhance apoptosis and cellular senescence. It likely regulates the abundance of myelin-specific sphingolipids galactosylceramide and sulfatide that affects myelin sheath architecture and motor neuron functions. It is located in the endoplasmic reticulum, and is a member of the ceramide synthase family (CerS1-6), with a broad distribution, particularly in kidney, liver, brain, heart, placenta and lung. It uses specific acyl-CoAs to generate ceramides essential for membrane structure and signaling. It acts primarily as a tumour suppressor by inhibiting metastasis, invasion, and proliferation in various cancers, including breast, liver, and gliomas.
