Antibody Host Species: Rabbit
Antibody Immunogen Source: Synthetic phosphopeptide patterned after human eIF4G1
Antibody Immunogen Sequence: CATKR(pS)F(pS)KEVE
Antibody Immunogen Description: Corresponds to amino acid residues A1181 to E1191. S1185 phosphorylation stimulates enzymatic activity. S1185 and S1187 are amongst the most phosphorylated sites with ≥187 and ≥82 mass spectrometry reports, respectively, in PhosphoSitePlus. eIF4G1 is known to be phosphorylated in vitro at S1185: by PKCA (PRKCA).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.
Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.
Antibody Concentration: 0.8 mg/ml
Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol
Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Scientific Background: eIF4G1 (Eukaryotic Translation Initiation Factor 4 Gamma 1) is a critical, large scaffolding protein that belongs to the eukaryotic initiation factor 4G family. eIF4G1 is vital for cellular growth, brain development, and is often overexpressed in cancers. It is essential for cap-dependent mRNA translation initiation that acts as a key component of the eIF4F complex. It binds eIF4E (cap-binding), eIF4A (helicase), and eIF3 (ribosome), and facilitates ribosome recruitment. It features distinct binding domains for eIF4E (N-terminal), eIF3/eIF4A (central), and a second eIF4A-binding site in the C-terminal. It exists in two complexes, either with EIF1 or with EIF4E. Together with EIF1, is required for leaky scanning, in particular for avoiding cap-proximal start codon. Together with EIF4E, it antagonizes the scanning promoted by EIF1-EIF4G1 and locates the start codon (through a TISU element) without scanning. It facilitates 5' UTR scanning, cap-dependent, and some cap-independent initiation processes, notably aiding translation during cellular stress. As a member of the eIF4F complex, it is required for endoplasmic reticulum stress-induced ATF4 mRNA translation. Deficiency of eIF4G1 is linked to impaired hippocampus-dependent learning, reduced mitochondrial energy production, and abnormal neuron morphology. It is overexpressed in various cancers (e.g., prostate, breast), driving tumour growth, survival, and invasion. This description may include information annotated by UniProt and/or Google AI.
