Antibody Immunogen Description: Corresponds to amino acid residues D59 to D66. The effect of S63 phosphorylation is unclear. This is the major in vivo site of phosphorylation of VAMP1 based on ≥22 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: VAMP1 (vesicle-associated membrane protein) is an 18 kDa member of the synaptobrevin family of proteins. It is an integral membrane protein crucial for neurotransmitter release and vesicle fusion at the presynaptic nerve terminal. It acts as a key component of the SNARE complex, mediating docking and exocytosis of synaptic vesicles. It participates in vesicle fusion with the plasma membrane (1). Synaptobrevins, Syntakins and the 25 kDa synaptosomal associated SNAP25 are the main components of a protein complex involved in the docking and/or fusion of synaptic vesicles with the presynaptic membrane (2). VAMP-1 is also one of the key proteins in the SNARE complex which is involved in regulated exocytosis (3). Primarily found in the nervous system (specifically brain and spinal cord) and neuromuscular junctions, with some isoforms expressed in other tissues (e.g., kidney, exocrine pancreas). Multiple splice variants exist, with VAMP1A (primarily neural) and VAMP1B (broadly expressed) being common. Mutations in VAMP1 are linked to serious disorders such as congenital myasthenic syndromes and hereditary spastic ataxia, resulting from weakened neurotransmitter release.
