Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: JNK1 (MAPK8) is a protein-serine/threonine kinase of the CMGC group and MAPK family. It is involved in the regulation of several cellular processes, including cell proliferation, differentiation, migration, transformation, and apoptosis, through the JNK/SAP signalling pathway. This pathway is activated by extracellular stimuli such as pro-inflammatory cytokines or physical stress, as well as signals of intracellular stress, such as misfolded proteins in the endoplasmic reticulum. It is also activated by high fat diet in fat, muscle, and liver tissues, leading to increased phosphorylation of IRS1 and contributing to insulin resistance. Phosphorylation of Thr-183 and Tyr-185 by either of two dual specificity kinases, MAP2K4 and MAP2K7, increases phosphotransferase activity. It is inhibited by dual specificity phosphatases, such as DUSP1, which target the T183 and Y185 activatory phosphorylation sites. It promotes apoptosis (cell death) by upregulating pro-apoptotic genes, but can also promote survival in specific contexts. Apart from c-Jun, substrates include proteins involved in T-cell differentiation, microtubule dynamics (via STMN2 phosphorylation), and the circadian clock.
