Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: p38d (MAPK13) is a protein-serine/threonine kinase of the CMGC group and MAPK family. It is essential for MAP kinase signalling that can be induced via physical stress or proinflammatory cytokines. p38d is activated by both proinflammatory cytokines (e.g., TNF-alpha, IL-1-beta), and chemical stress and environmental stress (UV radiation). p38b is directly activated by phosphorylation at Tyr-182 and probably Thr-180 by MKK3 (MAP2K3, MEK3) and MKK6 (MAP2K6, MEK6). p38d signalling can lead to the activation of the transcription factors ELK1 and ATF2, and other kinases such as MAPKAPK2. It can affect proliferation, differentiation, transcription regulation, translation regulation, cytoskeleton regulation, and development. Insulin secretion and pancreatic beta cell survival can be regulated by this kinase. p38d can be a positive or negative regulator of the above processes depending on the cell context. It has also been implicated in promoting heart hypertrophy. Increased expression of p38d has been associated with increases proliferation and metastatic progression. It is also hypermethylated in some cancer cell lines. The rare liver cancer Cholangiocarcinoma (CCA, CC) has been noted to have a significant up-regulation of p38d compared to normal bile duct tissue. It can promote cancer progression, particularly in breast cancer, where it contributes to tumour cell proliferation, and lung metastasis.
