Antibody Immunogen Description: Corresponds to amino acid residues G198 to A206. The effect of T202 phosphorylation is unclear, but this is located in the extra long activation loop in TTBK1 between subdomains VII (DGF) and VIII (GPE). Phosphorylation of this site has not been reported by mass spectrometry in PhosphoSitePlus. The site is highly conservered in animals, including sea urchin. The phosphosite sequence is also found in human TTBK2 surrounding T189.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: TTBK1 (Tau-tubulin kinase 1) is a neuron-specific serine/threonine and tyrosine kinase that belongs to the casein kinase-1 superfamily. This dual specificity kinase functions to phosphorylate tau protein, stabilizing microtubule assemblies, and its hyperphosphorylation activity is linked to tauopathies, such as Alzheimer's disease, where TTBK1 expression is elevated. It is crucial for tau phosphorylation, specifically at Ser-422, which induces tau aggregation. It may also play a role in synaptic vesicle formation by phosphorylating synaptic vesicle protein 2A (SV2A). TTBK1 is localized in the cytoplasm of neuronal cells. It features an N-terminal dual-kinase domain, a 39-amino acid polyglutamate stretch, and PEST sequences. It is expressed in the brain, particularly in cortical and hippocampal neurons, and weakly expressed in spinal cord and testis. Overexpression of TTBK1 is linked to impaired spatial learning, decreased motor function, and increased tau accumulation. It is also involved in M1-activated microglia-induced neurotoxicity.
