Antibody Immunogen Description: Corresponds to amino acid residues R230 to G236. The effect of T233 phosphorylation is unclear. This site has not been reported as phosphorylated in vivo as confirmed by mass spectrometry in PhosphoSitePlus. However, it is highly conserved and found in zebrafish.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ROCK1 (Rho-associated, coiled-coil-containing protein kinase 1; ROK-beta) is a 158. kDa protein-serine/threonine kinase of the AGC group and DMPK family. It is a key regulator of the actin cytoskeleton and cell polarity. It acts as a major downstream effector of the small GTPase RhoA, and is essential for regulating the actomyosin cytoskeleton, cell contractility, morphology, and motility. It is activated by binding the small G protein RhoA. ROCK1 regulates the cytoskeleton by phosphorylating targets like myosin light chain (MLC) and LIM kinase, which leads to increased contractility and stress fiber formation. It also plays a key role in metabolic stress-induced autophagy by phosphorylating Beclin1. ROCK1 features an N-terminus kinase domain and a C-terminus Rho-binding domain/pleckstrin homology domain. D1113A substitution leads to abolished cleavage by caspase-3. It is nvolved in cardiovascular diseases (e.g., hypertension), cancer metastasis, and fibrosis.
