Antibody Immunogen Description: Corresponds to amino acid residues R1049 to D1055. The effect of S1052 phosphorylation is unknown. This is the major phosphosite in NEK1 by mass spectrometry (≥112 reports recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Nek1 (NIMA-related kinase 1) is a protein serine/threonine protein kinase in the Other Group and NEK family. It also has phosphotransferase activity towards tyrosine residues. It is critical for DNA damage repair, cilia assembly, and cell cycle regulation. NEK1 is highly expressed in germ cells and plays a key role in meiosis. It is involved in DNA damage checkpoint control and for proper DNA damage repair, in part through Nek1 phosphorylation of VDAC1 to limit mitochondrial cell death. It acts in the early response to DNA damage, specifically in repairing double-strand breaks by stabilizing the ATR-ATRIP kinase complex, often operating independently of canonical ATM/ATR mechanisms. It features an N-terminal kinase domain (around residues 4–258) and a large C-terminal extension with coiled-coil regions that mediate protein-protein interactions. It regulates the assembly of primary cilia and maintains centrosome stability. Nek1 has been linked to polycystic kidney disease, short rib-polydactyly syndrome, short-rib thoracic dysplasia 6 with or without Polydactyly, asphyxiating thoracic dystrophy (ATD), sensenbrenner syndrome (CED), and short rib-polydactyly syndrome Type 2.
