Antibody Immunogen Description: Corresponds to amino acid residues H1407 to A1413. T1410 phosphorylation is stimulates enzymatic activity. This is a minor in vivo phosphorylation site in LRRK2 (≥4 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). LRRK2 is known to be phosphorylated at this site in vitro by LRRK2.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: LRRK2 (Leucine-rich repeat kinase 2; Dardarin) is a 268 kDa protein with multiple domains including several ankyrin, leucine-rich, and WD40 repeats, a Ras-like small GTPase family domain named ROC, and a MAPKKK kinase domain that is closely related to the RIP kinase domain. It acts as a signaling molecule and scaffold protein, phosphorylating a subgroup of Rab GTPases (e.g., Rab8A, Rab10) to regulate intracellular trafficking and lysosomal functions. LRRK2 positively regulates autophagy through a calcium-dependent activation of the CaMKK/AMPK signalling pathway. The process involves activation of nicotinic acid adenine dinucleotide phosphate (NAADP) receptors, increase in lysosomal pH, and calcium release from lysosomes. It is found in the cytoplasm and associated with membranes of organelles such as lysosomes, endosomes, and the Golgi apparatus. LRRK2 is expressed in brain as well as in other tissues such as lung, liver and heart. It is also highly expressed in immune cells, particularly neutrophils. LRRK2 might be central to the pathogenesis of several major neurodegenerative diseases associated with Parkinsonism. Mutations (e.g., G2019S, R1441G) typically cluster in the catalytic ROC-COR-Kinase core, increasing kinase activity.
