Antibody Immunogen Description: Corresponds to amino acid residues V228 to G234. T231 phosphorylation stimulates phosphotransferase activity. This phosphosite is located in the kinase activation loop between catalytic subdomains VII and VIII. This is a major site of in vivo phosphorylation based on ≥55 mass spectrometry reports recorded in PhosphoSitePlus. STLK3 is known to be phosphorylated at this site in vitro by STLK3, and Wnk1 (PRKWNK1).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: STLK3 (STK39; SPAK) is a protein-serine/threonine kinase in the STE Group, STE20 family and FRAY subfamily. It is a component of the WNK-SPAK/OSR1 kinase cascade, which is implicated in ion transport, and responses to hypertonic stress and blood pressure. It mediates the activation of the p38 MAP kinase pathway, and primarily functions as a mediator that phosphorylates cation-chloride-coupled cotransporters, affecting cell volume regulation and blood pressure control. It is phosphorylated by WNK1, WNK2, WNK3 or WNK4, and in turn phosphorylates ion cotransporters, such as SLC12A1/NKCC2, SLC12A2/NKCC1, SLC12A3/NCC, SLC12A5/KCC2 or SLC12A6/KCC3.
