Antibody Immunogen Description: Corresponds to amino acid residues G584 to F590. S587 phosphorylation appears to induce RNA splicing. This is a minor site of in vivo phosphorylation of SRPK1 based on ≥1 mass spectrometry reports recorded in PhosphoSitePlus. SRPK1 is known to be phosphorylated at this site in vitro by SRPK1.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SRPK1 (Serine-arginine protein kinase 1; SFRSK1) is a protein-serine/threonine kinase of the CMGC group and SRPK family. It is a highly conserved, constitutive enzyme that specifically phosphorylates serine-arginine (SR)-rich splicing factors (e.g., SRSF1 (ASF/SF2, SC35, and SRp55), essential for mRNA processing, and controlling their localization and activity in the nucleus. It plays an important role in regulating alternative splicing, cell survival, and tumour angiogenesis. It regulates angiogenesis (VEGF-A165/VEGF-A165b balance) and is involved in HIV-1 viral core phosphorylation. This kinase is moderate to highly expressed in most tested human tissues except apparently in the brain and spinal cord. It featuresf two kinase domains separated by a distinct spacer region. SRPK1 is activated by phosphorylation at Ser-51 and Ser-555. Elevated SRPK1 expression is linked to tumour growth, metastasis, and poor prognosis in many cancers by activating paths like PI3K/AKT, NF-κB, and Wnt/β-catenin. In human cancer cells, it was observed that inactivation of SRPK1 induces cellular resistance to anticancer drugs such as cisplatin and bleomycin.
