Antibody Immunogen Description: Corresponds to amino acid residues K329 to G239. The effect of their phosphoryation on RIOK2 is unknown. S332+S335+S337 are major sites of phosphorylation of RIOK2 from mass spectrometry studies (≥40-, >37-, and >50-times, respectively). RIOK2 is known to be phosphorylated in vitro at S335 by Plk1 (PLK).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: RIOK2 (RIO Kinase 2) is a protein-serine/threonine kinase in the Atypical Group, RIO family and RIO2 subfamily. It regulates the final steps of cytoplasmic maturation of the 40S ribosomal subunit by facillitating the export of the 40S pre-ribosome particles (pre-40S) from the nucleus to the cytoplasm. It provides for release of NOB1, PNO1 and LTV1 from the late pre-40S and the processing of 18S-E pre-rRNA to the mature 18S rRNA. It also regulates the timing of the metaphase-anaphase transition during mitotic progression, and its phosphorylation, most likely through PLK1.
