Antibody Immunogen Description: Corresponds to amino acid residues R140 to R146. The effect of S143 phosphorylation is unknown. S143 is the most common phosphosite in MYT1 by mass spectrometry (≥23 reports recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: MYT1 (PKMYT1) is a protein-serine/threonine kinase in the Other Group and WEE family. It serves as a negative regulator of entry into mitosis and meoisis (G2 to M transition) by phosphorylation of the CDK1 kinase at Thr-14 specifically when CDK1 is complexed to cyclins. To a lesser extent, it may directly or indirect also catalyze the phosphorylation of Tyr-15. Phosphorylation of CDK1 prevents premature mitosis, and is essential for Golgi/ER assembly during telophase. It is mainly found associated with the endoplasmic reticulum and Golgi complex, rather than in the nucleus. MYT1 is linked to Dysembryoplastic Neuroepithelial tumours (DNT, DNET), which are rare brain tumours that often affect the temporal lobe or the caudate nucleus.
