Antibody Immunogen Description: Corresponds to amino acid residues A663 to P670. The effect of T667 phosphorylation is unknown. It appears to be the most commonly observed phosphosite in GCN2 from high throughput mass spectrometry studies (≥22 reports recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: EIF2AK4 (eIF-2-alpha kinase GCN2) belongs to the protein kinase superfamily, Ser/Thr protein kinase family, GCN2 subfamily. It is a metabolic-stress sensing protein kinase that phosphorylates the alpha subunit of eukaryotic translation initiation factor 2 (eIF-2-alpha/EIF2S1) on 'Ser-52' in response to low amino acid availability. It functions as an activator of the integrated stress response (ISR) required for adapatation to amino acid starvation. It converts phosphorylated eIF-2-alpha/EIF2S1 into a competitive inhibitor of the translation initiation factor eIF-2B that results in repression of global protein synthesis and reduced overall utilization of amino acids. It also mediates translational initiation activation of specific mRNAs, such as the transcriptional activator ATF4, and permits ATF4-mediated reprogramming of amino acid biosynthetic gene expression to alleviate nutrient depletion. It plays a proapoptotic role in response to glucose deprivation. It promotes global cellular protein synthesis repression in response to UV irradiation independently of the stress-activated protein kinase/c-Jun N-terminal kinase (SAPK/JNK) and p38 MAPK signaling pathways. It binds uncharged tRNAs. EIF2AK4 is involved in cell cycle arrest by causing cyclin D1 mRNA translation repression after the unfolded protein response pathway (UPR) activation and cell cycle inhibitor CDKN1A/p21 mRNA translation activation in response to amino acid deprivation. It functions in the consolidation of synaptic plasticity, learning as well as formation of long-term memory, and plays a role in neurite outgrowth inhibition. It is also involved in the antiviral response against alphavirus infection by impairing early viral mRNA translation of the incoming genomic virus RNA, and preventing alphavirus replication. EIF2AK4 modulates the adaptive immune response to yellow fever virus infection, promotes dendritic cells to initiate autophagy and antigene presentation to both CD4(+) and CD8(+) T-cells under amino acid starvation.
