Antibody Immunogen Description: Corresponds to amino acid residues K232 to D238. The effect of Y235 phosphorylation is unclear. This is one of the minor in vivo phosphorylation sites in CaMK1a (≥2 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CaMK1a is a protein-serine/threonine kinase of the CAMK group and CAMK1 family. It plays a key role in mediating calcium signals to downstream targets, such as the transcription activator HDAC5 or the signalling protein MARK2. Phosphorylation at Thr-177 increases its phosphotransferase activity by approximately 25-fold in a Ca(2+)-CaM dependent manner. Thr-177 is phosphorylated by CAMKK1 or CAMKK2. It is essential for neuronal differentiation, dendritic spine growth, and the creation of long-term potentiation (LTP) in hippocampal neurons. It regulates axonal extension by affecting growth cone motility. It promotes neurite outgrowth, axon extension, and spine formation by activating targets like MARK2 and ARHGEF7. Often found in the cytoplasm and postsynaptic density, with roles in mediating NMDA receptor-induced calcium signalling.
