Antibody Immunogen Description: Corresponds to amino acid residues K171 to G177; in the protein kinase catalytic domain activation T loop region between subdomains VII and VIII. T174 phosphorylation is stimulatory for phosphotransferase activity. This is the major site of in vivo phosphorylation of YSK1 based on ≥158 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: YSK1 (Yeast Sps1/Ste20-related Kinase 1; STK25) is a protein-serine/threonine kinase that is oxidant stress activated and hypothesized to play a role in the cellular response to environmental stress. YSK1 is activated by the Golgi matrix protein GM130. It controls Golgi apparatus reorganization, which is essential for protein transport events, cell adhesion, and polarity complexes involved in cellular migration.It is part of the striatin-interacting phosphatase and kinase (STRIPAK) complexes, which regulate multiple pathways including Hippo signaling and cytoskeletal remodeling. YSK1 phosphorylates and activates ATG4B at Ser-383, which increases autophagic flux. It also plays a role in regulating apoptotic processes (cell death). Its C-terminal non-catalytic region inhibits YSK1's phosphotransferase activity. Unlike many other Ste20-related kinases, YSK1 lacks the Cdc42/Rac1-binding domain. It is ubiquitously expressed in various tissues but shows high expression in the brain and testis. Genetic inhibition of YSK1 has been shown to improve metabolic health in models of type 2 diabetes.
