Antibody Immunogen Description: Corresponds to amino acid residues D674 to K680. T676 and likely S677 phosphorylation stimulates phosphotransferase activity. Phosphorylation regulates cell cycle progression and molecular association. This phosphosite is located in the kinase activation loop between catalytic subdomains VII and VIII. This is one of the less major sites of in vivo phosphorylation of TTK (≥1 mass spectrometry report in PhosphoSitePlus). TTK is known to be phosphorylated at this site in vitro by TTK.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: TTK (ESK) is a protein-serine/threonine kinase of the Other group and TTK family. It is a dual specificity protein kinase that can phosphorylate serine, threonine, and tyrosine residues and has a role in cell cycle progression through regulating centrosome duplication during S-phase. This function was CDK2-mediated by direct activation of TTK.. Through CDCA8 phosphorylation TTK can modulate AurKB at the centromere to align the chromosomes. Phosphorylation of S582, T675, T676, T686, and S742 increases phosphotransferase activity. Phosphorylation of T806 inhibits phosphotransferase activity. TTK is a dual-specificity protein kinase with activity toward serine, threonine and tyrosine residues.
