Scientific Background: TIE2 (TEK) is a receptor protein-tyrosine kinase of the TK group and Tie family. It functions as a membrane receptor for ANGPT1, ANGPT2, and ANGPT4 and is involved in the regulation of angiogenesis, endothelial cell survival, proliferation, migration, adhesion, actin cytoskeleton dynamics, and vascular quiescence. TIE2 expression is required for the normal development of the heart and blood vessels and for hematopoiesis post-natally. After birth, TIE2 expression can either promote or inhibit angiogenesis dependent upon the cellular context. In addition, TIE2 also has anti-inflammatory activity by preventing pro-inflammatory plasma proteins and leukocytes from leaking through the blood vessel endothelium. TIE2 directly phosphorylates DOK2, GRB7, GRB14, PIK3R1, Shc1, and TIE1 to mediate its effects. In mice, rats, and humans, TIE2 expression is almost exclusively in endothelial cells that line the vasculature. TIE2 has been demonstrated to be essential for the maintenance of hematopoeitic stem cells (HSCs) in a quiescent state in the bone marrow, which is required for the ongoing process of hematopoeisis. Phosphorylation at Y897 inhibits TIE2 phosphotransferase activity.
