Antibody Immunogen Description: Corresponds to amino acid residues L160 to G166. T163 phosphorylation is predicted to stimulate phosphotransferase activity. This phosphosite is located in the kinase activation loop between catalytic subdomains VII and VIII. This is a major site of in vivo phosphorylation site of SIK3 based on ≥27 high throughput mass spectrometry reports recorded in PhosphoSitePlus. SIK3 is known to be phosphorylated at this site in vitro by LKB1 (STK11).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SIK3 (Salt-Inducible Kinase 3; QSK) is a 140 kDa protein-serine/threonine kinase of the CAMK group and CAMKL family. It is a member of the salt-inducible kinase family. It phosphorylates key substrates to manage energy storage, lipid metabolism, and cholesterol synthesis. It also positively regulates mTOR signaling by triggering the degradation of the inhibitor DEPTOR. SIK3 is crucial for homeostatic sleep/wake regulation, where a conserved phosphorylation site (Ser-551 in humans) regulates daily nonrapid-eye movement sleep amounts. It also controls the cell cycle by regulating HDAC4 trafficking. SIK3 is activated by phosphorylation on Thr-163 by STK11 in complex with STE20-related adapter-alpha (STRAD alpha) pseudo kinase and CAB39. It is a AMPK-family kinase that is ubiquitously expressed in the cytoplasm. It is widely expressed in tissues like the liver and brain.
