Antibody Immunogen Description: Corresponds to amino acid residues R355 to A361; After the kinase catalytic domain. S358 phosphorylation stimulates phosphotransferase activity. This is one of the major sites of in vivo phosphorylation of SIK2 based on ≥143 mass spectrometry reports recorded in PhosphoSitePlus. SIK2 is known to be phosphorylated at this site in vitro by Akt1 (PKBa), Akt2 (PKBb), PKACa (PRKACA), and SIK2 (QIK).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SIK2 (Salt-inducible kinase 2; QIK) is a 94 kDa protein-serine/threonine kinase of the CAMK group and CAMKL family. It is a member of the salt-inducible kinase family. It regulates insulin signaling, adipocyte differentiation, and fat/cholesterol synthesis, with high expression in skeletal muscle and adipose tissue. SIK2 may modulate the efficiency of insulin signal transduction, and affect insulin resistancy. It regulates autophagy when proteasomal function is compromised and interacts with p97/VCP to control endoplasmic reticulum-associated protein degradation (ERAD). It features a kinase domain at the N-terminal, a central SNF1-protein kinase homology (SNH) domain, and a phosphorylation domain near the C-terminal. It is activated by phosphorylation on Thr-175 by LKB1 (STK11) in complex with STE20-related adapter-alpha (STRAD alpha) pseudo kinase and CAB39. SIK2 kinase activity is regulated by an acetylation-deacetylation switch; p300/CBP-mediated acetylation at Lys-53 inhibits it, while HDAC6-mediated deacetylation restores it.
