Target Full Name: Tyrosine-protein kinase SgK223; Homologue of rat pragma of Rnd2

Target Alias: FLJ00269; SG223; Sugen kinase 223

Product Type Specific: SgK223 protein kinase phosphosite-specific antibody

Antibody Code: PK810

Antibody Target Type: Phosphosite-specific

Antibody Phosphosite: Y413

Protein UniProt: Q86YV5

Protein SigNET: Sgk223

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Human Sgk223 sequence peptide Cat. No.: PE-04APO99

Antibody Immunogen Sequence: EPI(pY)AES(bA)C

Antibody Immunogen Description: Corresponds to amino acid residues E410 to S416; in the first third of the protein. Y413 phosphorylation induces cytoskeletal reorganization and cell motility. This is the major in vivo site for phosphorylation of Sgk223 based on ≥714 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 1 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Potency: Very strong immunoreactivity with immunogen peptide on dot blots.

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Dog.

Antibody Positive Control: The observed molecular mass of the processed target protein on SDS-PAGE gels is reported to be around 150-170 kDa.

Scientific Background: SgK223 (Sugen kinase 223; NACK; Pragmin, serum-glucocorticoid regulated kinase-like (PRAG1)) is a protein-serine/threonine kinase of the Other group and NKF3 family. It appears to be a human pseudokinase and scaffolding protein that functions as an oncogenic driver in several cancers. It features an N-terminal region, a largely unstructured central PEST region (harboring phosphorylation sites), and a C-terminal pseudo-kinase domain (PsK). Its kinase-like domain is catalytically inactive and unable to bind ATP, and acts primarily by assembling signalling complexes that regulate cell morphology, motility, and invasion. It may regulate neurite outgrowth via activiation of RhoA (based on rat studies). SgK223 acts as an effector of Rnd2 (a Rho family GTPase) and promotes Src Family Kinase (SFK) signaling. It recruits C-terminal Src kinase (Csk) to the plasma membrane upon phosphorylation at Tyr-411, preventing Csk from turning off Src, which results in sustained SFK activation. It forms homo- and heterodimers with its structural homolog SgK269/PEAK1 to regulate signaling outputs.SgK223 is overexpressed in various cancers—including pancreatic, breast, and colon cancers—where it promotes cell migration and invasion.
