Scientific Background: RSK3 (RPS6KA2) is a protein-serine/threonine kinase of the AGC group and RSK family. It is a protein-serine/threonine kinase member of the RSK (ribosomal S6 kinase) family of growth factor-regulated kinases. RSK proteins have two kinase catalytic domains and are phosphorylated by MAPK proteins in response to growth factors, polypeptide hormones, and neurotransmitters. RSK3 is activated by multiple phosphorylations on threonine and serine residues. Phosphorylation at S360 increases phosphotransferase activity. Phosphorylation at S218 inhibits interaction with AKAP6. RSK3 acts downstream of ERK (MAPK1/ERK2 and MAPK3/ERK1) signalling and mediates mitogenic and stress-induced activation of transcription factors, regulates translation, and mediates cellular proliferation, survival, and differentiation. It may function as tumour suppressor in epithelial ovarian cancer cells.
