Antibody Immunogen Description: Corresponds to amino acid residues K214 to G221; in the protein kinase catalytic domain activation T loop region between subdomains VII and VIII. RSK3 S218 phosphorylation stimulates phosphotransferase activity and inhibits interaction with AKAP6. These are two of the major sites of in vivo phosphorylation of RSK2 based on ≥5 and ≥6 mass spectrometry reports recorded in PhosphoSitePlus. RSK3 is known to be phosphorylated in vitro at S218 by RSK3 (RPS6KA2).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: RSK3 (RPS6KA2) is a protein-serine/threonine kinase of the AGC group and RSK family. It is a protein-serine/threonine kinase member of the RSK (ribosomal S6 kinase) family of growth factor-regulated kinases. RSK proteins have two kinase catalytic domains and are phosphorylated by MAPK proteins in response to growth factors, polypeptide hormones, and neurotransmitters. RSK3 is activated by multiple phosphorylations on threonine and serine residues. Phosphorylation at S360 increases phosphotransferase activity. Phosphorylation at S218 inhibits interaction with AKAP6. RSK3 acts downstream of ERK (MAPK1/ERK2 and MAPK3/ERK1) signalling and mediates mitogenic and stress-induced activation of transcription factors, regulates translation, and mediates cellular proliferation, survival, and differentiation.
