Antibody Immunogen Description: Corresponds to amino acid residues N463 to V469; in the C-terminal third of the kinase after the catalytic domain. This is a major in vivo phosphorylation site in RIOK1. The effect of Y466 phosphorylation is unclear. This is a major in vivo phosphorylation site in RIOK1 based on ≥91 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: RIOK1 (RIO Kinase 1) is a protein-serine/threonine kinase of the Atypical group and RIO family. It is an ATPase, essential for 40S ribosomal subunit maturation, cell growth, and division. It plays a crucial role in ribosome biogenesis by facilitating the final cytoplasmic maturation of 40S ribosomal subunits (18S-E pre-rRNA to 18S rRNA). It features a central RIO domain, and is localized primarily to the cytosol. It acts as an ATPase to facilitate the release of biogenesis factors and is considered an atypical kinase because it acts mostly as an ATPase and not a classical kinase. It acts as a substrate adaptor for the PRMT5 complex, involved in arginine methylation. RIOK1 mediates p53 degradation, thus promoting cancer cell survival, and it supports Akt signaling and mTOR activation, controlling cell growth. It localizes to the nucleus. It exists as two alternatively spliced isoforms. RIOK1 is frequently overexpressed in several human cancers (including breast, lung, and prostate) and is linked to high tumour grade and poor survival rates.
