Antibody Immunogen Description: Corresponds to amino acid residues E577 to S583; In protein kinase catalytic domain activation T-loop between subdomains VII and VIII. Y579 and Y580 phosphorylation stimulate phosphotransferase activity. These are two of the major sites of in vivo phosphorylation of PYK2 based on ≥971 and ≥991 reports, respectively, from high throughtput mass spectrometry studies recorded in PhosphoSitePlus. PYK2 is known to be phosphorylated in vitro at Y579 by Src, and at Y580 by Src.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PYK2 (PTK2B, FAK2) is a cytoplasmic protein-tyrosine kinase of the TK group and Fak family. It is a non-receptor kinase that has a role in actin cytoskeleton rearrangement, cell migration, cell polarization, adhesion, bone remodeling, and controlling amount of marginal B-cells' migration and presence in the spleen. It is closely related to focal adhesion kinase (FAK). It acts as a bridge for Ca2+-dependent signals, linking extracellular stimuli to downstream pathways like MAPK to control cytoskeletal remodeling, cell migration, survival, and adhesion. It features an N-terminal FERM domain and a C-terminal focal adhesion targeting (FAT) domain. It is rapidly activated by increased intracellular Ca2+, PKC activation, or G-protein-coupled receptor activation. Upon activation, it autophosphorylates, creating docking sites for Src family kinases.
