Antibody Immunogen Sequence: SDI(pY)AEI(bA)C
Antibody Immunogen Description: Corresponds to amino acid residues S399 to I405; in the region between the FERM_M domain and the protein kinase catalytic domain. Y402 phosphorylation stimulates phosphotransferase activity. Its phosphorylation regulates cell cycle progression, cell motility, alters intracellular location, and induces binding to ErbB2, RASA1 and Src. This is one of the minor sites of in vivo phosphorylation of Pyk2 based on ≥25 mass spectrometry reports recorded in PhosphoSitePlus. PYK2 is known to be phosphorylated at this site in vitro by PYK2 (PTK2B, FAK2), and Src.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.
Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Scientific Background: PYK2 (PTK2B, FAK2) is a cytoplasmic protein-tyrosine kinase of the TK group and Fak family. It is a non-receptor kinase that has a role in actin cytoskeleton rearrangement, cell migration, cell polarization, adhesion, bone remodeling, and controlling amount of marginal B-cells' migration and presence in the spleen. It is closely related to focal adhesion kinase (FAK). It acts as a bridge for Ca2+-dependent signals, linking extracellular stimuli to downstream pathways like MAPK to control cytoskeletal remodeling, cell migration, survival, and adhesion. It features an N-terminal FERM domain and a C-terminal focal adhesion targeting (FAT) domain. It is rapidly activated by increased intracellular Ca2+, PKC activation, or G-protein-coupled receptor activation. Upon activation, it autophosphorylates, creating docking sites for Src family kinases. Phosphorylation at Tyr-402 increases phosphotransferase activity and induces interaction with ErbB2, RasGAP and Src. It Bbinds with Src family kinases, paxillin, and p130CAS, playing a critical role in scaffolding complexes. Phosphorylation at Tyr-579 increases its phosphotransferase activity. NPHP1 interaction with PYK2 has been interrupted with a P859A mutation. A phosphorylation site of PYK2 can be inhibited with an Y881F mutation, which will also impair GRB2 interaction. It is highly expressed in neurons, where it regulates NMDA receptor function, ion channels, and synaptic plasticity. It is. essential for macrophage and T/B cell signaling, chemotaxis, and adhesion. It regulates cell migration, spreading, and the disassembly of endothelial junctions. This description may include information annotated by UniProt and/or Google AI.
