Antibody Immunogen Description: Corresponds to amino acid residues V198 to G206; In protein kinase catalytic domain activation T-loop between subdomains VII and VIII. T201 and T203 phosphorylation are predicted to stimulate phosphotransferase activity. These are two of the major sites of in vivo phosphorylation of PRKX based on ≥12 and ≥28 mass spectrometry reports recorded in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PRKX (Protein Kinase X-linked; PKX1) is a protein-serine/threonine kinase of the AGC group and PKA family. It is an ancient, cAMP-dependent kinase that regulates crucial developmental processes, particularly renal epithelial morphogenesis and angiogenesis. Downstream targets of PRKX can include SMAD6, CREB, and PKD1. SMAD6 phosphorylation mediates differentiation of myeloid cells. It is expressed in various tissues, with the highest levels found in the fetal and adult brain, kidney, and lung. In the fetal kidney (specifically in the ureteric bud epithelium), PRKX acts to stimulate renal epithelial cell migration and branching morphogenesis. It stimulates crucial endothelial cell behaviours, including proliferation and the formation of vascular-like structures. While its catalytic core is highly conserved, its N-terminus is distinct from traditional PKA catalytic subunits. PRKX interacts with polycystin-1 and, when active, can rescue renal structural defects caused by PKD1 gene deficiency, indicating a role in mitigating Polycystic Kidney Disease (PKD) progression. It has been implicated in the tumorigenic process of nodular basal cell carcinoma and is part of the WNT signaling pathway.
