Antibody Immunogen Description: Corresponds to amino acid residues R238 to G244; in the protein kinase catalytic domain activation T loop region between subdomains VII and VIII. S241 phosphorylation is stimulatory for phosphotransferase activity. Phosphorylation induces interaction with 14-3-3 eta and 14-3-3 theta. This is the major in vivo phosphorylation site in PDK1 (≥334 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). PDK1 is known to be phosphorylated at this site in vitro by PDPK1 (PDK1).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: PDK1 (PDPK1) is a protein-serine/threonine kinase of the AGC group and PKB family. It features a catalytic kinase domain and a C-terminal pleckstrin homology (PH) domain. PDK1 activates, via phosphorylation, many members of the AGC family of protein kinases (e.g., Akt/PKB, PKC, S6K, and SGK). It regulates cell growth, survival, and proliferation. It is recruited to the plasma membrane by binding to phospholipids like phosphatidylinositol-(3,4,5)-trisphosphate (PIP3), which allows it to phosphorylate and activate substrates. PDK1 is activated by phosphorylaton at Tyr-9, Ser-160, Ser-241, Tyr-373, and Tyr-376. MELK-mediated phosphorylation can be inhibited through a T354A mutation. PDK1 phosphorylation of MAP3K5 can be ablated through the S394A and S398A mutations. PKB has been observed to be phosphorylated more via a T513E mutation in PDK1.
