Antibody Immunogen Description: Corresponds to amino acid residues F208 to G214; In protein kinase catalytic domain activation T-loop between subdomains VII and VIII. T211 phosphorylation stimulates phosphotransferase activity. This is a minor in vivo phosphorylation sites in Nuak1 (≥1 report from high throughput mass spectrometry studies recorded in PhosphoSitePlus). NuaK1 is known to be phosphorylated at this site in vitro by LKB1 (STK11), and NDR2 (STK38L).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: NuaK1 ((NUAK family SNF1-like kinase 1;ARK5) is a 74-76 kDa protein-serine/threonine kinase of the CAMK group and CAMKL family. It is a key member of the AMPK family involved in energy stress responses, and mediates cell proliferation, senescence, cell ploidy levels, and cell adhesion. It is Involved in tolerance to glucose starvation. It is activated by phosphorylation at Thr-211 by LKB1 and Ser-600 by Akt. A T211A NuaK1 mutation can inhibit LKB1/LKB1-mediated activation and phosphorylation of NuaK1, while ablating induction of senescence. The I400K and L401K mutations can lead to interrupted NuaK1 complex with PPP1CB, and inhibiting regulation of PP1 activity. It suppresses Fas-induced apoptosis by phosphorylation of CASP6, and suppresses the activation of the caspase and the subsequent cleavage of CFLAR. It phosphorylates ATM. Ploidy by NuaK1 is mediated through the phosphorylation of Ser-464 of LATS1. Cell adhesion control by NuaK1 is mediated via the Myosin Phosphatase 1 (PP1) complex and contributes to DNA repair by regulating p53 and CDKN1A (p21). Phosphorylation via PKB/Akt1 can be prevented through the S600A mutation. It functions as a tumor malignancy-associated factor, promoting survival under nutrient starvation and enabling cancer cell invasion and metastasis. Elevated levels of NUAK1 are associated with poor prognosis in various malignancies, including glioma and ovarian cancer.
