Antibody Immunogen Description: Corresponds to amino acid residues T191 to G198; in the protein kinase catalytic domain activation T loop region between subdomains VII and VIII. T191 and S195 phosphorylation is predicted to be stimulatory for phosphotransferase activity. T191 is a minor phosphosite (≥1 report) and S195 is a major phosphosite (≥80 reports) from high throughput mass spectrometry studies recorded in PhosphoSitePlus. Nek7 is known to be phosphorylated in vitro at S195 by NEK9.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Nek7 is a protein-serine/threonine kinase that is a member of the Other group of protein-serine/threonine kinases in the Nek family, NIMA subfamily. It plays a critical role in centrosome organization, microtubule nucleation, and cytokinesis, while also mediating inflammatory responses. It phosphorylates key proteins like Eg5 to facilitate centrosome separation before nuclear envelope breakdown. Beyond mitosis, NEK7 is a necessary activator of the NLRP3 inflammasome, which initiates inflammatory responses. It directly binds to NLRP3, allowing its oligomerization, independent of its kinase activity. Nek7 is highly expressed and widely distributed in most tested human tissues, including heart, brain, liver, testis, and leukocytes. In contrast to the other documented NimA-related kinases, Nek7 harbor its catalytic domain in the C-terminus of the protein. Immunofluorescence studies indicate that Nek7 is cytoplasmic. The major protein kinase that was active on the p70 S6 kinase hydrophobic regulatory site (FXXFS/TF/Y) Thr-412, was purified from rat liver and identified as Nek7. Its phosphotransferase activity is rapidly and efficiently increased by serum deprivation, and may be regulated in a cell cycle-dependent manner. Nek7 has been significantly associated with certain clinicopathologic indices in gallbladder carcinomas. The high level expression of Nek7, FoxM1 and Plk1 was significantly associated with shorter overall survival time in 76 specimens of gallbladder carcinomas, pericarcinomas and normal tissues (PubMEd: 23359173). High expression of NEK7 is linked to various cancers (e.g., ovarian serous carcinomas), and abnormal expression can lead to mitotic errors, such as micronuclei formation, cell cycle arrest, and death.
