Antibody Immunogen Description: Corresponds to amino acid residues A203 to G209. S206 phosphorylation stimulates phosphotransferase activity. This phosphosite is located in the kinase activation loop between catalytic subdomains VII and VIII. This is the major in vivo phosphorylation site in Nek6 (≥60 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). Nek6 is known to be phosphorylated at this site in vitro by NEK9.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Nek6 is a protein-serine/threonine kinase that is a member of the Other group of protein kinases in the Nek family. It is important for mitotic cell cycle progression, particularly in regulating chromosome segregation, robust spindle formation, and the metaphase-anaphase transition. It is a globular, monomeric protein that features a NIMA-like catalytic domain and a flexible N-terminal domain that mediates protein interactions. Nek6 is closely related in its catalytic domain to the protein-serine/threonine kinase NimA of Aspergillus nidulans. It is activated by Nek9, and phosphorylation by checkpoint kinases Chk1/Chk2. It phosphorylates proteins such as Eg5 (kinesin) for control of spindle assembly, as well as STAT3, Histone H3, and others. Nek6 is highly expressed and widely distributed in most tested human tissues except apparently in the brain and spinal cord. Nek6 is phosphorylated and activated during M phase of the cell cycle, and its inhibition facilitates apoptosis. Inhibition of Nek6 function, by either overexpression of an inactive Nek6 mutant or elimination of endogenous Nek6 by siRNA, arrests cells in M phase and triggers apoptosis. Nek6 may be an oncoprotein (OP). Nek6 may contribute to tumorigenesis by suppressing p53/TP53-induced cancer cell senescence. It is often overexpressed in cancer cells (breast, liver, prostate, etc.), where it aids tumour progression by suppressing p53-induced cellular senescence.
