Antibody Immunogen Description: Corresponds to amino acid residues K747 to Y754; In protein kinase catalytic domain activation T-loop between subdomains VII and VIII. Y749 and Y753 phosphorylation stimulate phosphotransferase activity. This is one of the major in vivo phosphorylation sites in MERTK (≥449 and ≥403 reports, respectively, from high throughput mass spectrometry studies recorded in PhosphoSitePlus). MERTK is known to be phosphorylated in vitro at Y749 by MER (MERTK), and at Y753 by MER (MERTK).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: MERTK (MER) is a protein-tyrosine kinase of the TK group and Axl family. It is a receptor kinase that activates downstream signaling (e.g., PI3K/Akt, MAPK/ERK) upon binding ligands such as Gas6, Tubby, or Galectin-3 . It functions in the regulation of cell survival, angiogenesis, migration and invasion, differentiation, and promotes non-inflammatory phagocytosis of apoptotic cells and suppresses inflammation. It is critical for phagocytosis of rod outer segments by the retinal pigment epithelium (RPE). It features two Ig-like domains and two FNIII domains, and anchored in the plasma membrane with a transmembrane domain. Phosphorylation of Tyr-749, Tyr-753 and Tyr-754 increases phosphotransferase activity. It is highly expressed in macrophages, dendritic cells, NK cells, and epithelial cells.
