Antibody Immunogen Description: Corresponds to amino acid residues R236 to D242; in the region between the PB1 domain and the protein kinase catalytic domain. The effect of S239 phosphorylation is unclear. This is a major site of phosphorylation for MEKK2 (≥281 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: MEKK2 (MAP3K2) is a 70 kDa protein-serine/threonine kinase of the STE group and STE11 family. It functions as a critical MAP3K in signaling pathways regulating immune response, inflammation, and cell proliferation. It acts as an upstream kinase, primarily activating the JNK1/2, p38, ERK5, and NF-κB pathways, but It also binds to Protein Kinase C-related Kinase 2 (PRK2). It is required for cytokine production in cultured mast cells. The protein is one of the kinases that regulate different steps of the caveolar cycle. It features a C-terminal catalytic serine/threonine kinase domain and a distinct N-terminal regulatory domain. MEKK2 activity is regulated through 14-3-3 protein binding, which modulates dimerization. It is also activated by phosphorylation on Ser-514, Ser-520, and Thr-522. It is also known to form a complex with Mip1, which acts as an inhibitor. Methylation of the protein increases MAP kinase signalling and promotes Ras-driven carcinomas. Overexpression of MEKK2 has been associated with colorectal cancer progression, where it typically resides in the cytoplasm.
