Antibody Host Species: Rabbit
Antibody Immunogen Source: Human MAPKAPK5 (PRAK) sequence peptide Cat. No.: PE-04AAS95
Antibody Immunogen Sequence: PQF(pT)PYYV(bA)C
Antibody Immunogen Description: Corresponds to amino acid residues P183 to V190; in the protein kinase catalytic domain activation T loop region between subdomains VII and VIII. T186 phosphorylation is predicted to be stimulatory for phosphotransferase activity. This is one of the less predominant in vivo phosphorylation sites in MAPKAPK5 (≥11 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.
Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.
Antibody Concentration: 0.5 mg/ml
Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol
Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Scientific Background: MAPKAPK5 is a protein-serine/threonine kinase of the CAMK group and MAPKAPK family. It is activated by MAPK1/ERK, p38 kinases a/b, or ERK3/4 following phosphorylation of Thr-182, translocating from the nucleus to the cytoplasm to regulate cell proliferation and senescence. It is activated by stress-related extracellular stimuli; such as H2O2, arsenite, anisomycin TNF alpha and also PMA and the calcium ionophore A23187; but to a lesser extent. In vitro, it is activated by SQSTM1. T182A substitution leads to no p38-beta/MAPK11-induced activation. In addition, T182D, S212D and L337G mutations are associated with higher protein kinase activity. It appears to be a tumour suppressor protein (TSP) that mediates Ras-induced senescence and phosphorylates p53/TP53. It also phosphorylates FOXO3, ERK3/MAPK6, ERK4/MAPK4, HSP27/HSPB1, and RHEB. It inhibits mTORC1 signaling by phosphorylating RHEB. MAPKAPK5 is expressed ubiquitously in human tissues. This description may include information annotated by UniProt and/or Google AI.
