Antibody Immunogen Description: Corresponds to amino acid residues N949 to K955; in the C-terminus of the kinase. The effect of T952 phosphorylation is unclear. This is the major in vivo phosphorylation site in LOK (≥112 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). LOK is known to be phosphorylated at this site in vitro by ERK1 (MAPK3), and ERK2 (MAPK1).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: LOK (Lymphocyte-Oriented Kinase; STK10) is a 130-138 kDa protein-serine/threonine kinase of the STE group and STE20 family. It can control lymphocyte migration via its phosphotransferase activity. LOK autophosphorylates and phosphorylates meylin basic protein, histone 2A, and MSN (moesin). It is similar to Polo-like kinase kinases and can associate with and phosphorylate PLK1 to affect cell cycle progression. LOK can negatively regulate MEKK1/MAP3K1. It is abundant at the lymphocyte plasma membrane, where it is critical for cytoskeletal rearrangement and other T-cell responses. It features a kinase catalytic domain at the N-terminus (STE20-like) and a long coiled-coil structure at the C-terminus. LOK is highly expressed in lymphocytes, with significant levels in the spleen, thymus, and peripheral blood leukocytes. A K65I mutation in LOK seen with Testicular germ cell tumour (TGCT) can induce loss of kinase phosphotransferase activity.
