Antibody Immunogen Description: Corresponds to amino acid residues R425 to K431; in the C-terminus of the kinase. S428 phosphorylation stimulates phosphotransferase activity. Phosphorylation regulates cell growth, intracellular location and molecular association, including inducing interaction with STRAD and exportin 1. This is one of the minor in vivo phosphorylation sites in LKB1 (≥5 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). LKB1 is known to be phosphorylated at this site in vitro by ATM, DNAPK (PRKDC), LKB1 (STK11), MSK1 (RPS6KA5), p70S6K (RPS6KB1), PKACa (PRKACA), PKCz (PRKCZ), and RSK1 (RPS6KA1).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: LKB1 (STK11) is a protein-serine/threonine kinase of the CAMK group and CAMKL family. LKB1 has important roles in the regulation of cell metabolism, cell polarity, apoptosis, and the response to DNA damage. LKB1 promotes recruitment of p53/TP53 to the CDKN1A/WAF1 promoter to activate transcription. in addition, LKB1 regulates p53/TP53-dependent apotosis pathways. The LKB1 protein is found in the nucleus and cytoplasm of cells and translocates to the mitochondria during apoptosis. LKB1 is activated by binding of a complex consisting of CAB39 and STRAD or CAB39 and ALS2CR2. Phosphorylation at S307 induces interaction with AMPK1, STLK5 and exportin 1. Phosphorylation at Ser-428 increases phosphotransferase activity. Phosphorylation at Thr-336 inhibits LKB1.
