Antibody Immunogen Description: Corresponds to amino acid residues P487 to H493; in the region between the Ksr1-SAM and kinase catalytic domains. The effect of S490 phosphorylation is unclear. This is the major in vivo phosphorylation site in Krs2 (≥48 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Ksr2 (kinase suppressor of Ras 2) is a protein-serine/threonine kinase of the TKL group and RAF family. It is a scaffold protein that regulates essential signaling pathways, particularly the Raf-MEK-ERK cascade, playing a critical role in energy homeostasis, fatty acid oxidation, and glucose metabolism. The Ksr2 protein has very low kinase catalytic activity, but it can phosphorylate MAP2K1 in vitro with low efficiency at several serine and threonine residues. Ksr2 functions to inhibit MAP3K3-mediated activation of ERK, JNK, and NF-kappa-B signalling pathways, thus blocking MAP3K-mediated production of interleukin-8 (IL-8). KSR2 interacts with calcineurin as a substrate to modulate calcium-dependent ERK activation. In addition, Ksr2 interacts with AMPK (the cellular energy sensor) to maintain energy homeostasis, cellular thermogenesis, fat oxidation, and glucose utilization. Mutations in KSR2 are linked to early-onset obesity, hyperphagia (excessive hunger), and insulin resistance.
