Scientific Background: Ksr2 (kinase suppressor of Ras 2) is a protein-serine/threonine kinase of the TKL group and RAF family. It functions as a scaffold protein that connects MEK and Raf isoforms. The Ksr2 protein has very low kinase catalytic activity, but it can phosphorylate MAP2K1 in vitro with low efficiency at several serine and threonine residues. Ksr2 functions to inhibit MAP3K3-mediated activation of ERK, JNK, and NF-kappa-B signalling pathways, thus blocking MAP3K-mediated production of interleukin-8 (IL-8). In addition, Ksr2 interacts with AMPK (the cellular energy sensor) to maintain energy homeostasis and glucose utilization.
